Stay one night in the middle, that's Yosemite Valley, but you have to book in advance. I booked all the rooms for the end of September in early April. If you can't get a room, stay in West Yosemite; leave San Francisco early in the morning and you can arrive at Yosemite before noon. You can play for half a day without hiking. On the second day, drive to Las Vegas. It's estimated that you'll be driving for a whole day.

My route is Salt Lake City - Arches National Park - Las Vegas. The drive from Arches to Las Vegas took 7-8 hours, with both of us taking turns driving and no accommodation in the middle.
